چکیده

Polymerase Chain Reaction (PCR) is the principal method of amplifying target DNA regions and, as such, is of great importance when performing microbial diversity studies. An unfortunate side e↵ect of PCR is the formation of unwanted byproducts such as chimeras. The main goal of the work covered in this article is the development of an algorithm that simulates realistic chimeras for use in the evaluation of chimera detection software and for investigations into the accuracy of community structure analyses. Experimental data has helped to identify factors which may cause the formation of chimeras and has provided evidence of how influential these factors can be. This article makes use of some of this evidence in order to build a model with which to simulate the PCR process. This model helps to better explain the formation of chimeras and is therefore able to provide aid to future studies that intend to use PCR.

Kinetic Study and Modelling of n-Heptane Reforming Process over Pt/Zr-HMS/HZSM-5 Composite Catalysts

Pt/Zr(x)-HMS/HZSM-5 catalysts with various Si/Zr ratios (as x) were used for investigation of the kinetics of n-heptane reforming process under 350 - 450 oC reaction temperature, 35 to 80 ml min-1 flowing rate of hydrogen and 2 to 5 ml h-1 flowing rate of n-heptane. In the present work, two kinetics (power law and Langmuir - Hinshelwood) models were selected and examined to describe the kinetic...

A Novel Integrated Approach to Modelling of Depletion-Induced Change in Full Permeability Tensor of Naturally Fractured Reservoirs

More than half of all hydrocarbon reservoirs are Naturally Fractured Reservoirs (NFRs), in which production forecasting is a complicated function of fluid flow in a fracture-matrix system. Modelling of fluid flow in NFRs is challenging due to formation heterogeneity and anisotropy. Stress sensitivity and depletion effect on already-complex reservoir permeability add to the sophistication. Horiz...

Micelle PCR reduces chimera formation in 16S rRNA profiling of complex microbial DNA mixtures

16S rRNA gene profiling has revolutionized the field of microbial ecology. Many researchers in various fields have embraced this technology to investigate bacterial compositions of samples derived from many different ecosystems. However, it is important to acknowledge the current limitations and drawbacks of 16S rRNA gene profiling. Although sample handling, DNA extraction methods and the choic...

Chip Formation Process using Finite Element Simulation “Influence of Cutting Speed Variation”

The main aim of this paper is to study the material removal phenomenon using the finite element method (FEM) analysis for orthogonal cutting, and the impact of cutting speed variation on the chip formation, stress and plastic deformation. We have explored different constitutive models describing the tool-workpiece interaction. The Johnson-Cook constitutive model with damage initiation and damag...

Mathematical Modelling of Pulmonary Edema

The excess accumulation of water in lung interstitial or alveolar is called pulmonary edema which is caused by factors that upset the normal Starling balance in micro-circulation. Pulmonary edema disturbs the alveolar gas exchanges which are normally regulated by the respiratory system. Mathematical modelling of pulmonary edema may help to predict the lung conditions and the mechanisms involved...
